After you arrive at the airport which is 5km west of the city on highway 3 you have 2 ways to get to the centre of Yellowknife. You can take a taxi which would cost about 14$ and you can get one by calling 867/873-4444 or you can take a bus via Fort Smith for 100$. I think the first is better :).
