The Brandon bus terminal has many Greyhound buses that are running to Winnipeg, Regina and Saskatoon as well as other places in southwest Manitoba so this would be the best place to go according to your needs. You can call and buy tickets ahead at 204/727-0643 or 1-800/661-8748. You also have Grey Goose Liners that run buses on Highway 10.
